Los efectos saludables de los ácidos grasos poliinsaturados omega-3 (AGPI-n3), EPA y DHA, hacen que las industrias farmacéutica y alimentaria tengan un gran interés en la investigación y producción de estos concentrados. Monoglicéridos y diglicéridos de AGPI-n3 son altamente biodisponibles y estables a la oxidación lipídica. La reacción de glicerolisis catalizada por lipasas es la mejor alternativa para producir estos concentrados. Para solventar el problema de inmiscibilidad entre los sustratos, aceite de sardina y el glicerol, se han evaluado diferentes estrategias, como son el uso de tert-alcoholes, surfactantes, CO2 supercrítico y un proceso de pre-emulsificación de los sustratos. En relación al estado de oxidación lipídica, se ha comprobado que las lipasas inmovilizadas adsorben los productos de oxidación formados durante la reacción de glicerolisis. Además, también se ha optimizado un proceso de destilación molecular para fraccionar los productos de reacción y concentrar los MAG ricos en AGPI-n3.
Production of omega-3 polyunsaturated fatty acids (n-3 PUFA) concentrates is of great interest for pharmaceutical and food industries. It is well known that the main n-3 PUFA, EPA and DHA, have important health benefits. n-3 PUFA monoacylglycerides and diacylglycerides (n-3 MAG and DAG) present good bioavailability and oxidation stability. The lipase-catalyzed glycerolysis reaction is the best alternative to produce the n-3 MAG. The main drawback of this process is the immiscibility of the substrates. The use of tert-alcohols, surfactants, supercritical CO2 and a substrate emulsification process have been evaluated to improve the contact between sardine oil and glycerol to achieve high yields of the reaction products. Regarding oxidation state, immobilized commercial lipases have presented adsorption capacity of the oxidation products formed during the glycerolysis reaction. Furthermore, molecular distillation process has been also optimized to fractionate the reaction products and concentrate the MAG rich in n-3 PUFA.
