In additionto the main proteins of this signaling pathway, there are severalregulatory proteins known as scaffold proteins, that modulate theintensity, amplitude and duration of ERK signals. Interestingly, it canpropose the scaffold proteins as antitumoral targets. Considering thehypothesis that scaffold proteins interact and regulate the signaltransduction through the Ras-ERK pathway in a coordinated mannerwe try to find out how these interactions are regulated. Ras proteinsare distributed in different types of plasma membrane: microdomainsand endomembranes. We have previously demonstrated thatcompartmentalization dictates Ras utilization of effectors, the intensityof its signals and the biological response of the cells. Cell migration iscritical for many physiological processes and is often misregulated indevelopmental disorders and pathological conditions including cancer.MAPK signaling are known regulators of cytoskeletal dynamics duringcellular processes such as cell adhesion and migration. Our hypothesisis that localized ERK signaling mediated through recently identifiedscaffold proteins and its interactions with the cytoskeleton mayregulate specific cellular responses. Here we propose that associationsamong different scaffold proteins and cytoskeleton will add one furtherdegree of regulation for an already tightly regulated cascade and couldprovide a novel means for manipulating ERK signals, even withtherapeutic purposes.
